Gene Synthesis

Gene synthesis is a brand new service from contractexpress that utilises proprietary technologies recently licensed from Gene Oracle. Customers simply provide an amino-acid or DNA sequence, and then we apply expert knowledge to optimize the construction of your gene to ensure efficient cloning and optimal protein expression. Once you are happy with the modifications we suggest, the fast and reliable synthesis process commences. Within 10 working days we will deliver your fully sequence verified gene returned to you in a cloning vector of your choice.

The contractexpress gene synthesis service has been developed for all scientists whether in academia, biotech or pharma, who need to:

  • Rapidly access the gene(s) for their research and development targets, especially where no gene clones are currently available.

  • Have the sequence of their gene(s) optimised to promote expression of the protein in their desired expression system.

  • Go further and combine gene synthesis with the contractexpress expression & purification services to evaluate expression of your gene(s) in bacterial or insect cells and to provide you with the purified protein(s) required.

Codon Optimisation

Each gene can be optimised to the codon usage table of specific cell types e.g. E. coli, CHO, Spodoptera.

The sequence is optimised to provide a balanced usage of codons in line with the specific cell type - this is to be distinguished from the use of the most abundant codons throughout the optimised gene. This has several potentially beneficial effects;

  • Avoids depletion of tRNA pool for abundant codons and hence potential protein expression problems.
  • Avoids the possibility of excessively rapid translation (through use of only abundant codons) leading to protein expression problems i.e. uncontrolled translation may lead to improper folding and expression machinery clogging.
  • Avoids the insertion of repeat codons into optimised sequences that may make them difficult to clone subsequently or even make the DNA unstable.

Cloning

Cloning is provided into the following vector(s) at no additional cost:

  • pDrive (Qiagen).

Charges for cloning will be applied if the user would like the gene to be cloned into a non-standard vector. However, these costs will be applied for the first time only and will not be applied if the customer requires the same vector to be used in subsequent contracts.

This vector can be supplied by the customer. If sourced by NextGen Sciences, the customer will be charged for the additional cost of NextGen Sciences obtaining the vector.


Delivery

Within 10 working days from receipt of order and deposit of the desired gene sequence through the secure website or via e-mail (for larger genes and for mutagenesis this may be longer but will be advised in advance).

Deliverables:

  • Plasmid DNA corresponding to cloned gene(s). Approx 1.5µg of DNA.
  • Agar stab containing cloned gene in E.coli lab strain e.g. NEB Turbo.
  • Sequence map of cloned gene in Genbank format.
  • Sequencing data confirming correct gene synthesis.
